DEVELOPMENT OF A MOLECULAR MARKER FOR RUST RESISTANCE GENES Sr39 AND Lr35 IN WHEAT BREEDING LINES Electron. J. Biotechnol.
Polymorphic DNA bands were identified between a near iso-isogenic line of wheat carrying both stem (Sr39) and leaf (Lr35) rust resistance genes and the recurrent line Thatcher (Tc) which lacks these genes. Both resistance genes are located on a translocated chromosomal segment derived from Aegilops speltoides and thus are genetically linked. The primers used to generate polymorphic bands were 3'-anchored inter-simple sequence repeat primers which identified genomic microsatellites with a repeated motif of 3 nucleotides in length. Expression of the antibody 14D9 in Nicotiana tabacum hairy roots Electron. J. Biotechnol.

Nicotiana tabacum hairy roots that express the antibody 14D9 were established. The 14D9 antibody yield obtained after 20 days of culture was 5.95 μg 14D9ml-1. The addition of the reticulum endoplasmic retention sequence KDEL demonstrated a positive effect over the intracellular 14D9 amounts with a yield increase up to 20.82 µg ml-1. DMSO increased the antibody amount in the biomass from 20.00 to 64.03 µg ml-1 while PVP (at 1.5 gl-1) and gelatine (at 5.0 gl-1) increased total 14D9 amounts in the culture medium to 25 µg and 14 µg respectively

Recombinant expression and refolding of the c-type lysozyme from Spodoptera litura in E. coli Electron. J. Biotechnol.

The chicken-type lysozyme of the insect Spodoptera litura (SLLyz) is a polypeptide of 121 amino acids containing four disulfide bridges and 17 rare codons and participates in innate defense as an anti-bacterial enzyme. The recombinant S. litura lysozyme (rSLLyz) expressed as a C-terminal fusion protein with glutathione S-transferase (GST) in Rosetta(DE3) Singles. The protein was produced as an inclusion body which was solubilized in 8 M urea, renatured by on-column refolding, and purified by reversed-phase chromatography to 95% purity. Background Angelica sinensis is a well-known traditional Chinese medicinal plant. We aimed to assess the genetic diversity and relationships in A. sinensis cultivars collected from different locations of China and also some other Angelica species. Results We employed an improved random amplified polymorphic DNA (RAPD) technique for the amplification of DNA materials from ten Angelica cultivars, and the results were verified by inter-simple sequence repeat (ISSR) analysis. Enzymatic synthesis of short chain citronellyl esters by a new lipase from Rhizopus sp Electron. J. Biotechnol.

Short chain citronellyl esters were synthesized by a new microbial lipase from Rhizopus sp strain isolated and lipase produced at UNICAMP, Brazil. Direct esterification and transesterification reactions have been performed to produce citronellyl acetate and butyrate in a free-solvent system and with n-hexane in reaction medium. Reaction mixture for direct esterification and transesterification was carried out at 45ºC in equimolar concentration of substrates: acid or ester and alcohol. Immobilized lysozyme protein on fibrous medium: Preliminary results for microfiltration applications Electron. J. Biotechnol.

The protein lysozyme was deposited onto a permeable support comprising chemically functionalized glass fiber. The main objective of this study was to set a stable organic net with no effect on the medium bed permeability and a preliminary test the activity of this enzyme under immobilized conditions. The influences of water activity and solvent hydrophobicity on the kinetics of the lipase-catalyzed enantioselective esterification of flurbiprofen with n-butanol were investigated. The solvent effect was not similar for lipases from Candida rugosa (Crl), Mucor javanicus (Mjl), and porcine pancreas (Ppl). The lipase-catalyzed reaction rates in different solvents across a wide range of water activities revealed that the Ppl-catalyzed reaction exhibited no enantioselectivity and no substantial water activity or solvent dependence. The Plackett-Burman design was used in the first step to evaluate the effects of eight factors, including six fermentation medium components and two cell adsorption conditions. A preferable approach to clone hLIF cDNA from the genomic DNA Electron. J. Biotechnol.

Complementary DNA (cDNA) is valuable for investigating protein structure and function in the research of life science, but it is difficult to obtain by traditional reverse transcription. In this study, we employed a novel strategy to clone the human leukemia inhibitory factor (hLIF) gene cDNA from genomic DNA directly isolated from the mucous membrane of mouth. The hLIF sequence can be acquired within a few hours by means of amplification of each exon and splicing using overlap-PCR. Thus, the new approach developed in this study is simple, time- and cost-effective, and it is not limited to particular gene expression levels of each tissue.

In the present report, citric acid production from raw glycerol in two fed-batch systems by acetate negative-mutants of Yarrowia lipolytica: Wratislavia 1.31 and Wratislavia AWG7 was compared. In the system, in which the total glycerol concentration was 200 g∙L-1, the substrate was added by pulsed additions, and in the other, in which the total glycerol concentration was 300 g∙L-1, the substrate was added at a constant feeding rate of 1.4 g∙h-1. Influence of metal ions and organic carbons on denitrification activity of the halotolerant bacterium, Paracoccus pantotrophus P16 a strain from shrimp pond Electron. J. Biotechnol.

The effect of metal ions, ferric ion (Fe3+) and molybdenum ion (Mo6+) on the denitrification process of Paracoccus pantotrophus P16 grown under saline conditions was investigated. Results revealed that the dosages of added Fe3+ and Mo6+ significantly accelerated nitrate utilization and nitrite accumulation.
